In order to ensure a sufficiently fast build-up of drug concentration at the site of action, there must be a correspondingly large concentration gradient between drug depot in the connective tissue and the endoneural space.
Accordingly, disintegration and dissolution will take place in the duodenum at normal speed (A, track 1) and drug liberation per se is not retarded. The liberation of drug, hence the site and time-course of absorption, are subject to modification by appropriate production methods for matrix-type tablets, coated tablets, and capsules.

 

Inhibition of the enzyme causes free NE concentrations to rise. Likewise, dopamine catabolism is impaired, making more of it available for NE synthesis. Consequently, the amount of NE stored in granular vesicles will increase, and with it the amount of amine released per nerve impulse. In the CNS, inhibition of MAO affects neuronal storage not only of NE but also of dopamine and serotonin.
The resultant wafers are remarkably robust, yet dissolve in very small quantities of water in seconds. This approach is especially attractive when working with highly potent or hazardous materials because it avoids manufacturing operations (granulation, blending, and tableting) that generate large quantities of aerosolized powder and present much higher risk of exposure.
